As soon as you have been scheduled an opponent for Day 2. That means you're next accepted challenge will be played on Day 3.
Good question - Added to the FAQ at the bottom of the OP.

Defending Champion LoL look strong
#1 LoL start off the season 2-0 coming into the first day of games in the Early-Season Tournament. LoL has 2 scorers averaging well over 20ppg already and will be the team to beat. Unfortunately they will have to wait a day before kicking off their campaign in the Early-Season Tournament. NFL Blitz, Manchester Rain, Animal House and The Magic Johnsons have also started 2-0. carl_johnson has to be happy with how his teams are performing and will look to avenge last season by coming out with a win in the Early-Season Tournament to stake his championship credentials.
On the other side of the ledger, Pacific Rimmers, Old School All-Stars, Team Wafu, Pimpology2 all stand at 0-2. Team Wafu and Old School All-Stars will face off tomorrow in the first day of the Early Season Tournament with the loser being consigned to an 0-3 start to their season. Meanwhile Pacific Rimmers will have to face the 2-0 Manchester Rain in what will be a tough matchup to start off with.
The game of the night will be NFL Blitz (2-0) vs The Magic Johnsons (2-0) with the winner of that game shaping up to be the early pick for Tournament Champion. Mayor of London, Boris Johnson has been doing it all for The Magic Johnsons so far with 15.5ppg, 8rpg, 4spg, 2.5bpg and while NFL Blitz continues to have All-Pro rebounders, it is Arizona Cardinals Cornerback Patrick Peterson that has been a real surprise with 8ppg, 7rpg, 7.5pbg over the first two games. The battle between those seven footers will certainly be a key part of the game.
The scheduled games for the first round (Day 3) in the Early Season Tournament is as below:
LoL; No game
Pacific Rimmers challenges Manchester Rain
Pimpology2 challenges CR Veterans
Team Wafu challenges Old School All-Stars
Old West challenges Animal House
NFL Blitz challenges The Magic Johnsons
Challenges may now be sent for Day 4: wrote:

That's the great thing about this league, both of these teams were set up horribly last season and now I get to start over and try again. Nevertheless, I don't expect any early season success, though, if I had kept Oasis' plan from last season, it probably would have been a winning strategy for the early season tournament.

The real question is, when did Boris Johnson and Patrick Peterson become seven footers? It's not every day you see seven-foot mayors or cornerbacks. Maybe if I keep the Magic Johnsons next season I'll make all real people have their actual height.

Agreed. It definitely makes me more interested in the regular season, even if my team plans won't reflect that.

NFL Blitz grab favourite tag
In the game of the night, NFL Blitz showed up and handily defeated The Magic Johnsons in what was a night of blowouts. Patrick Peterson dominated the much anticipated matchup with Boris Johnson, almost netting himself a double-double. Pimpology, Wafu and Animal House all scored blowout wins - But the tight game was between Manchester Rain and the Pacific Rimmers. Rimmers held the lead for almost the entire game before Manchester went on a 12-2 run in the last 4.47. The Rimmers will be given a day to cool off as they have a bye tomorrow, while Manchester Rain will look to extend their winning streak against defending West League champions LoL. Old School All-Stars join the Rimmers at 0-3 for the season - They might be able to grab their first scalp against CR Veterans, who will also be looking to return to form.
Tomorrow's game of the night is Manchester Rain (3-0) vs LoL (3-0). In LoL's first game of the tournament, they will be looking to take charge of the pool and remind everyone why they are defending champions. Oasis vocalist Liam Gallagher and Miami Heat superstar LeBron James will spearhead the scoring attack against LoL as they take up the challenge of going against premier rebounder/blocker . Alistar from LoL.
The following challenges for Day 5's games may now be sent:
LoL challenges Pacific Rimmers
